Interferon-α (IFN-α) preactivation recapitulates a Sjögren-like B cell phenotype. (A) fluorescence-activated cell sorter (FACS) sorted CD19⁺IgD⁺ cells from healthy donors (n=5) were cultured with IFN-α for 18 hours prior to stimulation with CpG+interleukin (IL)-10, imiquimod (Imiq)+IL-10, IL-10 alone or medium. Frequencies of CD19^dimCD138⁺ plasmablasts, and total IgM and IgG concentrations in supernatants were analysed at day 8. Data represent pooled data from 3 independent experiments. (B) Gene expression of cells was measured at day 0 and 8 by NanoString. Graph depicts values of mean fold change of clusters 1–14 in cells cultured for 8 days and normalised to expression day 0.
