(A) Live cell population (dual negative population for Annexin V and 7-AAD) after indicated dose of QC treatment in hDFs and hESCs (B) After 2 hours treatment with H₂O₂ or QC, hDFs (upper panels) or hESCs (lower panels) were stained with DCF-DA and ROS level was determined by flow cytometry. The relative level of DCF-DA positive population was shown as a bar graph (right panel). (C) hESCs were incubated with lucigenin with H₂O₂ or QC for 20 minutes. Relative level of ROS measured by luminometer was shown as bar graph. (D) Mitochondrial ROS production was determined by MitoSOX staining and following flow cytometry. The results represent one of the experiments performed twice.
