GC-1 spg cells were treated with melatonin of 10nM for 2 hrs, 6 hrs, and 12 hrs. The level of ERK1/2 phosphorylation was increased after melatonin treatment and detected by Western blot (A). Cells were treated ERK1/2 signaling inhibitor (PD98059, S1805, Beyotime, 30μM) for 2 hrs, then followed by melatonin treatment for 24 hrs. PD98059 inhibited the melatonin-induced proliferation of GC-1 spg cells (B). The qRT-PCR showed that Mt2 expression induced by melatonin was also decreased after PD98059 treatment (C). Different letters indicate significant differences (p < 0.05).
