(A, B) A172 cells were treated with EGF (10 ng/ml) in the presence or absence of LY294002 (10 μM) or U0126 (10 μM) for 48 h. (A) Expression levels of mRNA and protein and the activity of MMP-9 were determined by qRT-PCR, Western blotting, and zymography, respectively. GAPDH and MMP-11 served as internal controls. (B) Phosphorylation of ERK1/2, AKT, STAT3, and STAT5 were determined by Western blotting. (C, D) In some experiments, A172 cells were treated with EGF (10 ng/ml) in the presence or absence of STAT3 or STAT5 siRNA. The expression levels of mRNA and protein and the activity of MMP-9 then were determined.
