Trp73 deficiency impairs reprogramming efficiency, even in the absence of p53. MEFs of the indicated genotypes, cultured and treated identically, were transfected with OSKM (a and b) or OSK factors (c and d) and the reprogramming efficiency was monitored by quantification of alkaline phosphatase positive colonies (AP+) after either 22 days for WT and p73KO (a and b) or 17 days for p53KO and DKO (c and d) of doxycycline treatment. Representative scanned plates and photomicrographs (10 ×) of the colonies are shown for each condition. Two independent reprogramming experiments were performed, including at least three biological replicates from the indicated genotypes (with the exception of p53KO-MEFs, n=2), with two independent transfections per replicate.
