Cx43 forward trafficking and Cx43-20 K expression is regulated by mTOR signaling pathway in human myometrial cells. Representative images of hTERT-HMA/B cells transfected with Cx43-GFP and induced for PRA (a,b,c), or PRB (d,e,f) expression for 24 h and stimulated with P4 (100 nM) for 2 h. Results showed that Cx43 forward trafficking is promoted in PRA cells (a) and restricted in the PRB cells (d) however inhibition of mTOR signaling pathway via specific inhibitor PP242 (5 μM) resulted in the rescue of Cx43 trafficking defect in PRB cells (e) whereas stimulation of mTOR signaling pathway via insulin (10 μg/ml) treatment resulted in the inhibition of Cx43 trafficking in PRA cells (c).
