PU-H71 reduces CLL survival through the induction of mitochondrial apoptosis. (a) CLL cells were treated with PU-H71 (0–1 μM) for 48 h or 0.5 μM for 24–96 h. Apoptosis was measured by Annexin V/7-Aminoactinomycin D double staining. One of six cases was shown. (b) Cytochrome C release in PU-H71-treated CLL cells. Three of six cases are shown. Time and concentrations are indicated. (c) Western blot analysis of anti-apoptotic proteins in cells treated with PU-H71 at indicated concentrations for 48 h (N =6). (d) Western blot analysis of anti-apoptotic proteins in cells treated with 0.5 mM of PU-H71 for 24 or 48 h.
