Glutamate clearance is altered in Cln3 −/− astrocytes. The ability of wild type (WT) and Cln3-deficient (Cln3 −/−) astrocytes to clear glutamate from the medium was assessed using a Glutamate Assay Kit. WT and Cln3 −/− astrocytes were incubated with 2 mM glutamate for 2 h, and wells without astrocytes were used as controls. The glutamate remaining in the medium was quantified and normalized to the total amount of protein, and the glutamate uptake values of Cln3 −/− astrocytes were normalized to those of WT astrocyte samples. Cln3 −/− astrocytes took up significantly less glutamate than did WT astrocytes over the 2 h period
