Effects of CSZ on caspase-3 and -9 activitives in Aβ-stimulated SH-SY5Y cells. Caspase-3 and -9 activities were measured using each peptide substrate (DEVD-AFC and LEHD-AFC). (A) Caspase-3 activity in SH-SY5Y cells treated with Aβ (1–10 μM) or CSZ (1, 2.5 μM) for 20 h. (B) Caspase-3 activity was determined in SH-SY5Y cells incubated with or without 1 h pretreatment with CSZ or inhibitor of MEK1/2 or p38 MAPK followed by 20 h incubation with Aβ. Caspase-3 activity of the control cells was 60.54 ± 4.35 nmol/mg protein/h. (C) Caspase-9 activity in SH-SY5Y cells treated with Aβ (1–10 μM) or CSZ (1, 2.5 μM) for 20 h. (D) Caspase-9 activity was determined in SH-SY5Y cells incubated with or without 1 h pretreatment with CSZ or inhibitor of MEK1/2 or p38 MAPK followed by 20 h incubation with Aβ. Caspase-9 activity of the control cells was 25.72 ± 3.2 nmol/mg protein/h.
