Neutralization of secreted ASAH1 and Sph-1P with Anti-ASAH1 or Anti-Sph-1P antibodies resulted in reduced cell growth. Cells were grown in 96-well plates with serum-free media over a period of 48 h, after which MTT assays were performed to measure cell growth. U87-10gy cells were treated with 3 µg of either anti-ASAH1 (A) or humanized anti-Sph-1P (LT1009) antibodies (B). PBS, and/or 3 µg of non-specific IgG (NS), either control mouse or humanized IgG, were used as negative controls. Results shown are mean ± SD of three independent experiments. *P<0.05, compared with PBS and NS (non-specific IgG) groups.
