Mechanism of action of NR4A2 inhibitors. NR4A2 are regulated by a number of signaling pathways including downstream cascades of EGFR and Src. EGFR and Src inhibitors can suppress NR4A2 level via depleting p-ERK and possibly others such as PI3K, whereas individual downstream inhibitors (e.g., mTOR/S6K inhibitors) cannot do so. KU171309 seems to have a novel, unknown mechanism to downregulate NR4A2.
