Effect of proteins on exosome association. PC3-derived exosomes were treated with proteinase K to remove surface protein moieties. PC3-derived exosomes (untreated or subjected to proteinase K) were incubated with PC3 and MCF-7 cells for 4 hrs at 37°C. Association was quantified by monitoring the percentage of cells that had increased fluorescence when compared to control cells (“Positive”). For both PC3 and MCF-7 cells, untreated PC3 exosomes were associated with their cellular targets to a significantly greater extent than PC3 exosomes subjected to proteinase K (p<.05).
