Plasmids of the effector LD-VP and GD-PRE6 or GD alone as a control, and the reporter LexA:Gal4:GUS (diagrammed on the top) plasmids were co-transfected into protoplasts isolated from rosette leaves of 3 to 4-week-old Col wild type plants, the protoplasts were incubated for 20–22 h at room temperature in darkness, and GUS activity was measured.
