OSM induces DExD/H‐Box RNA helicase and IRF expression in human dermal fibroblasts (HDF). HDF were treated with OSM (20 ng/ml) for indicated time periods. Total RNA was prepared and subjected to reverse transcription. (A) IRF1, (B) DDX58, (C) IFIH1, (D) IRF7 and (E) IRF9 mRNA levels were analysed by quantitative real‐time RT‐PCR. Relative mRNA levels were normalized to the housekeeping gene, GAPDH, and fold changes were calculated relative to untreated sample (set to 1). Shown are the means (n = 4–5) with standard error of mean (S.E.M.). Statistical significance was assessed by unpaired t‐test. *P ≤ 0.05, **P ≤ 0.01, ***P ≤ 0.001 when compared to untreated sample. (F) Whole cellular lysates were prepared after stimulation with OSM (20 ng/ml) for the indicated time periods and expression of MDA5, RIG‐I and IRF1 was determined by Western blot analysis using specific antibodies. Tubulin was included as loading control. Blots shown are representative for three experiments.
