The release of LXA₄ is reduced in Anaplasma phagocytophilum-infected PMN. Primary human neutrophil granulocytes were coincubated with cell-free A. phagocytophilum for 300 min and extracellular bacteria were removed by washing. During the last 90 min of infection 10 ng/ml GM-CSF was added. For transcellular lipoxin synthesis A. phagocytophilum-infected and uninfected neutrophils were coincubated with HT-29 cells and stimulated with ionomycin (1.0 μM) for 10 (a) or 360 (b) min. LXA₄ content of the supernatants was determined by ELISA. n = 3 each, n.d.: not detectable, and ^∗p ≤ 0.05.
