GNA regulates the expression of miR-21 via inhibition of HIF-1α accumulation and STAT3 phosphorylation. (A) U266 cells were exposed to hypoxia in the presence or absence of GNA for 24 h. The expressions of HIF-1α, STAT3, p-STAT3 were analyzed by western blots. Data are presented as mean ± SD (n = 3). *P < 0.05 compared to hypoxia alone group. U266 cells were exposed to hypoxia in the presence or absence of (2-ME) or (S3I-201) for 24 h. (B) The levels of miR-21 were detected by qRT-PCR. (C) 2-ME reduced the HIF-1α protein expression and increased the PTEN protein expression. (D) S3I-201 reduced the p-STAT3 protein expression and increased the PTEN protein expression. Data are presented as mean ± SD (n = 3). *P < 0.05 compared to hypoxia alone group.
