Hsa-miR-25-3p inhibited endogenous CYP2B6 expression in HepaRG cells. Differentiated HepaRG cells were transiently transfected using 25 nmol/L miRNA negative controls or transfected with mimics or inhibitors for hsa-miR-25-3p and hsa-miR-504-5p. Together with the increased miRNA levels (A), decreased CYP2B6 mRNA levels (B) were observed in cells transfected with hsa-miR-25-3p, but not hsa-miR-504-5p. (C) Western blots show that CYP2B6 protein levels were decreased in HepaRG cells transfected with the hsa-miR-25-3p mimic but were increased in cells transfected with the hsa-miR-25-3p inhibitor.
