(A) Intracellular ROS level in C4-2AT6 cell was determined by fluorogenic marker carboxy-H2DCFCA. Fluorescence microscope detected elevated signals of GFP in C4-2AT6 cells treated with cabazitaxel (10 nM) than docetaxel (10 nM). CBZ: cabazitaxel, DOC: docetaxel. (B) Carboxy-H2DCFCA was quantified by FACS. FACS analysis revealed that treatment with docetaxel slightly increased ROS generation in C4-2AT6 cells. The fluorescent intensities were quantified by FACS. (C) Immunocytostaining showed significantly higher level of phosphor-p38MAPK in C4-2AT6 cell when treated with cabazitaxel, compared with docetaxel.
