Dynamic proteomics to study virus-host interactions in single cells. (A) Schematic representation of the screen. A CFP-expressing HSV-1 was allowed to adsorb to clones seeded in 12-well plates for 30 min and washed out, and then the cells were imaged every 20 min for 12 h. Overall, more than 50,000 single cells from 400 different YFP-expressing clones were monitored. (B) An example of a cellular protein whose level correlates with successful initiation of gene expression from the viral genome. RFX7 (yellow) levels are highly heterogeneous in the cell population.
