Determination of the absolute dissociation constants for Na+ (KD(Na+)) and H+ (KD(H+)). The apparent Na+-binding dissociation constants (Kd(Na+)) in the absence or presence of melibiose in the pH range from 5.55 to 8.45 were determined by ITC at 25°C. 1–20 mM NaCl was placed in the syringe. All apparent Kd(Na+) data were plotted against the H+ concentration without averaging and fitted by a linear function; the absolute KD(Na+) value corresponds to the y-axis intercept, and the absolute KD(H+) value corresponds to the x-axis intercept (see Materials and methods). pKa = −log KD(H+).
