Western blot analysis to determine NS5A domain bound by R9-HuscFvs. SDS-PAGE-separated GST-tagged-recombinant D1, D2, and D3 of NS5A and GST were probed with E-tagged R9-HuscFvs, followed by rabbit anti-E-Tag, goat anti-rabbit isotype-HRP conjugate and substrate, respectively. N, negative binding controls (antigen-blotted strips were incubated with buffer instead of the respective R9-HuscFv). P, positive controls (antigen-blotted strips were probed with goat-anti-GST). All of the R9-HuscFvs bound to the NS5A D1 and did not react to the D2 and D3 or GST control.
