HRP concentration dependency of the absorbance at 500 nm, A500, measured after 10 min incubation at 25 °C for [PPD] = 1.5 mM, [H2O2] = 80 μM and pH 7.0 (buffer-1). Each data point represents the average and standard deviation for three measurements using the same HRP and H2O2 stock solutions and a freshly prepared PPD stock solution. Linear regression of the data points yields a slope of 0.00144 pM−1, r2 = 0.9993. With ε500 = 1.109 × 104 M−1 cm−1 (see text), A500 (l = 1 cm) = 0.10 corresponds to 9.0 μM Bandrowski’s base.
