Emodin-induced CD4⁺CD25⁺ or CD8⁺CD122⁺ regulatory T cells (Tregs) suppress skin allograft rejection in Rag1^−/− recipient mice. CD4⁺CD25⁺ or CD8⁺CD122⁺ Tregs were isolated via FACS cell sorting from B6 recipient mice that were transplanted with BALB/c skin and treated with emodin and/or CsA. CD4⁺CD25⁺ (A) or CD8⁺CD122⁺ (B) Tregs (0.4 × 10⁶), together with B6-derived naïve CD3⁺ T cells (2 × 10⁶), were adoptively transferred to Rag1^−/− mice (B6 background) that were then transplanted with a BALB/c skin graft. Skin allograft rejection in Rag1^−/− recipient mice was observed (n = 7–8 grafts).
