FAM19A5 regulates RANKL-induced gene expression during osteoclastogenesis. (A,C,D) Mouse BMDMs were stimulated with FAM19A5 (2 μM) in the presence of M-CSF (30 ng/ml) and RANKL (100 ng/ml) for 3 days. Cells were harvested for RNA preparation. qPCR was performed using specific primers for RANK, TRAF6, OSCAR, TRAP, Blimp1, c-FOS, NFATc1, MafB, IRF-8, DC-STAMP, OC-STAMP, Atp6v0d2, and GAPDH. (B) Cells were harvested and Western blot analysis was conducted using anti-RANK, anti-TRAF6, anti-c-fos, anti-NFATc1, and β-actin antibodies. Data are presented as means ± SE (n = 3).
