(A) 293T cells were transfected with a plasmid that expresses FLAG-tagged NP encoded by influenza A 141/H1N1, WSN/H1N1, an H6N1 virus, or an empty vector. The lysates of transfected cells were subjected to anti-FLAG immunoprecipitation. The precipitates were separated by SDS-PAGE, and the gels were then subjected to silver staining. The indicated band was excised, digested, and subjected to MS for protein identification, as described in the materials and methods. (B) Lysates of 293T cells transfected with the indicated FLAG-tagged NP expressing plasmids were collected and subjected to anti-FLAG immunoprecipitation. The proteins in the lysates and precipitates were analyzed by SDS-PAGE and immunoblotting with anti-FLAG, anti-hnRNP A2/B1 and anti-β actin antibodies.
