Priming does not increase M‐CSF/IL‐4 BMMs’ ability to respond to TDB. BMMs were differentiated with GM‐CSF and M‐CSF/IL‐4 from C57BL/6 bone marrow over 8 days, primed with 0.5 ng/mL LPS for 24 h and then stimulated with 40 or 100 µg/mL TDB, or 100 ng/mL LPS as positive control. Mincle expression (A) was measured at 24 h using flow cytometry, and production of IL‐1β (B), IL‐6 (C), and TNFα (D) measured by ELISA at 48 h.
