Schematic diagram of the proposed fidelity checking process in DNA polymerase I. Following nucleotide insertion, the DNA is translocated to the checking site. If the inserted base pair is a correct pair, the polymerase continues. If not, the DNA is shifted to the exonuclease domain and the incorrect base pair is excised.
