Basolateral targeting determinant is located within the extreme COOH terminus of Kir 2.3. Reconstructed z-plane confocal images of MDCK cell monolayers stably transfected with various Kir 2.3 constructs. In contrast to the basolateral membrane expression of the wild-type (Wt) channel, mutant Kir 2.3 channels lacking the last 10–15 aa (435X, 431X) are expressed on the apical membrane (arrowhead), consistent with the removal of a basolateral membrane-targeting signal.
