Oligonucleotide substrates have been optimized for sequence and length so that reactions do not require RPA protein. (A) In the presence of RAD51 and ATP, radiolabeled homologous duplex and tailed duplex substrates are converted into heteroduplex and ssDNA products. Tailed duplex (63/32-mer) was constructed from oligos 5/6, and homologous duplex (31/31-mer) was constructed from oligos 7/8 (Table 1). (B) Results of reactions performed at 30ºC. (C) Results of reactions performed at 37ºC. Error bars in panels (B) and (C) represent results from 3 identical experiments. The signal from a background control reaction containing no protein was subtracted from all data.
