(A) Bioinformatics analysis showed the predicted binding sites between XIST and miR-139-5p. (B) Luciferase reporter assay demonstrated that miR-139-5p overexpression significantly decreased the luciferase activity of XIST-WT in HEK293T cells. (C) XIST knockdown increased the relative expression of miR-139-5p in T24 and 5637 cells. (D) There was no obvious change in XIST expression after transfection with miR-139-5p mimics in T24 and 5637 cells. (E) XIST could pull-down miR-139-5p expression in HEK293T cells by RNA pull-down assay. (F) The relative expression levels of miR-139-5p was significantly down-regulated in bladder cancer tissues compared to adjacent normal tissues. (G) The relative expression levels of miR-139-5p was significantly down-regulated in bladder cancer cell lines compared to normal urothelial cells.
