HCT-116 cells (A-C) or the primary-cultured human colon cancer cells (“Con Can1”, D-F) were treated with LB-100 at 10 μM, cells were further cultured for the designated time; Relative PP2A activity (A and D) was analyzed; Expressions of listed proteins were tested by Western blotting assay (B, C, E and F). Stable HCT-116 cells, expressing miR-17-92 expression vector (“miR-17-92-Vec”, three lines, “Line1/2/3”), non-sense control microRNA (“miR”), or the parental control cells (“Ctrl”) were subjected to quantitative real-time PCR (“qRT-PCR”) assay of miR-17-92 (G) and PP2A B subunit (H) as well as the Western blotting assay of listed proteins (I); Cell survival at 96 hours was tested by MTT assay (J).
