(A) Cultured BMDCs from Nlrp3A350V Tnf–/– mice had significantly reduced expression of pro-Casp1 and pro-Il1b mRNA and similar levels for Nlrp3, Asc, and pro-Il18 when compared with expression levels in Nlrp3A350V Cre-ERT2 BMDCs. (B) No significant differences were detected in the expression levels of inflammasome components or pro-cytokines between Nlrp3A350V Cre-ERT2 and Nlrp3A350V Tnf–/– BDMCs after stimulation with LPS (1 ng/ml). n = 5 mice per group for all measured values. *P < 0.05, by 2-way ANOVA with Dunn’s multiple comparisons test.
