Binding of daptomycin (2 μM) to POPC:POPG 80:20, containing 0.1 mol% LRh-DOPE vesicles (GUVs) in the presence of 2 mM Ca2+. A, All channels: daptomycin (cyan), carboxyfluorescein (green), and LRh-DOPE (red). B, Daptomycin channel only (cyan). Eleven trials were performed on vesicles composed of POPC:POPG 80:20 or 70:30 POPC:POPG containing 0.1 mol% LRh-DOPE in the presence of daptomycin and either in the absence or presence of Ca2+. Most solid red vesicles are multilamellar vesicles (or are out of the focal plane), which were excluded from the analysis. But some are GUVs with large inner vesicles, which were included.
