A., In-vitro CD4+ and CD8+ T-cell proliferation assays. CD4+ and CD8+ T-cells isolated from WT Balb/c mice were co-cultured with irradiated APCs and various concentrations of soluble anti-CD3 and OX40L-FP. H3-thymidine uptake was measured after 4 days in culture. B., Treg suppression assay. CD4+ responder cells from Balb/c mice were co-cultured with varying amounts of syngeneic Tregs, and stimulated with soluble anti-CD3, 10 µg/mL OX40L-FP, and irradiated APCs. H3-thymidine uptake was measured 3 days later.
