Proposed model for the role of focal adhesion kinase (FAK) in the metastasis of human epidermal growth factor receptor-2 (HER-2)-positive breast cancer cells. HER-2/FAK increases the phosphorylation of FAK/cSrc/p130Cas signaling. The phosphorylation of FAK reduces the expression of the epithelial maker, E-cadherin, and increases the expression of the mesenchymal markers, vimentin and fibronectin, the leading processes of epithelial-mesenchymal transition (EMT). The aforementioned processes promote cell migration and invasion to facilitate the metastasisof breast cancer.
