MiR‐130a directly targets PPARγ. A, An alignment between PPARγ and miR‐130a as predicted by TargetScan 6.2 analysis in various animals as shown by vertical bars. The sequence alignment of putative miR‐130a and its targeting site on 3′‐UTR of PPARγ shows a high level of complementarity and sequence conservation among vertebrates. B and C, HEK293 cells were transfected with 100 ng pEGFP, pEGFP‐PPARγ UTR or pEGFP‐PPARγ UTR mut along with control siRNA or miR‐130a mimetic, respectively. After 48 h of transfection, the EGFP gene expression (B) and protein level (C) were determined by qRT‐PCR and Western blotting, respectively. Data are expressed as means±SE from 3 independent experiments. **P<0.01 vs control siRNA. miR‐130a indicates microRNA‐130a; pEGFP, plasmid carrying enhanced green fluorescent protein; PPARγ, peroxisome proliferator‐activated receptor γ level; qRT‐PCR, quantitative reverse transcriptase polymerase chain reaction.
