Knockout of HuR reduces GM-CSF mRNA in CD4+ T cells. Naive WT and HuR KO CD4+ T cells were isolated as described in Materials and Methods and cultured with anti-CD3 and anti-CD28 stimulation in Th0 and Th17 cell polarizing conditions. Expression of GM-CSF mRNA (a,b) and protein (c–e) in Th0 and Th17 cells was measured by qRT-PCR, flow cytometry, and ELISA, respectively. Knockout of HuR reduced the GM-CSF mRNA and protein levels in Th0 and Th17 cells compared with WT counterparts. Results in (a,b,d and e) represent the summary of three independent experiments.
