PTEN-L and lclPTEN-L Designs

The top sequence shows the naturally occurring sequence coding for PTEN-L. The middle panel shows the sequence changes made to this naturally occurring PTEN-L sequence to generate the construct for high-level expression of PTEN-L. The methionine codon at the start of normal PTEN was also changed to a glycine codon in this construct to suppress expression of normal PTEN. The bottom sequence shows the sequence of lclPTEN-L in which the putative leader sequence is replaced by a human light-chain IgG leader sequence. Changed amino acids and bases with respect to the naturally occurring PTEN-L sequence are highlighted in yellow. Amino acids of the PTEN-L putative leader sequence and the human light-chain IgG leader sequence are in boldface.
