Assay for Cell-to-Cell Transfer of Catalytically Active PTEN

(A) Schematic of experiment. U87MG cells engineered for inducible lclPTENL were cocultured with U87MG cells expressing mCherry fluorescent protein. Cells with inducible lclPTENL are shown in white; cells expressing mCherry are shown in red. The motility of both populations is determined by videomicroscopy. (B–E) Cells expressing PTEN (B), lclPTEN-L (C), lclPTEN-L(C124S) (D), and PTEN-L (E) were cocultured with U87MG cells labeled with mCherry. Videomicroscopy was used to determine distance traveled for both cell populations in the cocultures in the absence and presence of doxycycline.
