Figure 2

Newcastle disease virus (NDV) mini-genome expression of nucleoprotein (NP) mutants. The NDV mini-genome expression of full-length NDV NP was compared with NP truncation mutants (A,B) and NP deletion mutants (C,D). (A and C) Schematic representation of full-length NDV NP-Myc and NP-Myc mutants. (B and D) Mini-genome RNA synthesis activity of the NP mutants. BSR T7 cells were transfected with plasmids as described in the Materials and Methods. The relative luciferase expression levels of cells transfected with plasmids encoding NP-Myc were defined as 100%. Expression of NP-Myc and NP-Myc mutants was detected via western blotting using an anti-Myc antibody. The gels are uncropped images. Asterisks indicate a statistically significant difference between the titers of the chimeric virus and the parental virus. P-values were calculated based on a two-tailed, unpaired t-test (95% confidence levels). *P = 0.01–0.05; **P = 0.001–0.01; ***P = 0.0001–0.001. n = 3.
