Effects of neutralizing TNFα antibody on the mRNA expressions of eNOS (A), gp91phox (B) and p22phox (C) in HUVECs. The conditioned medium was pre-incubated with neutralizing TNFα antibody for one hour to block TNFα effects, the medium then was added to HUVECs to incubate for another 24 hours, the mRNA expressions of eNOS, gp91phox and p22phox in HUVECs were determined by real-time PCR. TNFα antibody in the conditioned medium from nicotine-obese rats partially restored the expression of eNOS, gp91phox, p22phox. TNFα concentration in conditioned medium (D) or in plasma (F) was increased in obese (HFD) rats, and was further increased in nicotine-obese rats. *p<0.05 vs. vehicle medium; #p<0.05 vs. HFD; †p<0.05 vs. correspondence group without TNFα antibody.
