(B) ϕC31 integrase (400 nM) was added to pϕC31LRX substrate and the mixture was incubated at room temperature for 5 min to allow binding and full occupation of the sites; then gp3 (lane 3), ϕC31.Int(S12A)-gp3 (lane 4), or Bxb1.Int-gp3 (lane 5) was added. The mixture was then incubated for 1 h at 30°C, and the products were detected as in Figure 3.
