The presence of eHsp90 (left side), initiates signaling events that stimulate Src activation, leading to downstream molecular events such as RhoA activation, MLC2 phosphorylation, and assembly of EphA2-myosin complexes. In the context of eHsp90 and ephrin A1 ligand, these molecular events promote acto-myosin assembly and subsequent cell rounding, repulsion and cell-ECM detachment. The targeting of eHsp90 (right side), attenuates the Src-RhoA axis, preventing EphA2-myosin assembly, thereby interfering with ligand dependent cell rounding. In this scenario, cells remain morphologically flat and spread due to ECM adhesive forces. EphA2 internalization is not appreciably affected by eHsp90.
