DY1 is a chloroplast stroma protein. (A) Subcellular localization of DY1 in chloroplast. Protoplasts transformed with pA7-eYFP-DY1 were observed under a FLUOVIEW FV1000 Laser Confocal Microscopy System (Olympus) (Bars = 20 μm). (B) Western blot analysis demonstrating the stroma localization of DY1. Purified chloroplast were sub-fractionated into the stroma and thylakoid membrane fractions, separated by SDS–PAGE, blotting and probed with the antiserum against DY1. LHCB1 and RbcL were probed as controls. The bands of processed and mature forms of DY1 at 42 and 24 kDa, respectively, are indicated.
