MnP treatment reduces NF-κB binding in the liver. (A) After 12 weeks of HFD feeding, nuclear liver extracts from HFD and HFD + MnP mice were mixed with 32P-labeled NF-κB consensus oligonucleotide for Electrophoretic Mobility Shift Assay (EMSA). Control samples included 32P-labeled NF-κB probe alone, 32P-labeled NF-κB probe mixed with HeLa extract, and HeLa extract mixed with 50-fold excess unlabeled NF-κB consensus oligonucleotide. Samples were resolved on a gel. The image displays the NF-κB shift, non-specific binding (NS), and free probe; (B) Quantification of NF-κB binding was measured by densitometry.
