S100A7 outcompetes Zincon for Zn(II). Representative plots showing the response of 20 μM Zincon to Zn(II) in the presence of 10 μM S100A7 ox, S100A7-Ser, S100A7-Ala or S100A7Δox in 75 mM HEPES, 100 mM NaCl, pH 7.0, and 25 °C. The Zn(II)-Zincon complex exhibits an absorbance maximum at 621 nm. Data from titrations performed in the presence of Ca(II) and with the disulfide-null variants are presented in Figure S14.
