Figure 4.
Knockdown of SHISA2 expression by siRNA decreases PC cell numbers. (A) siRNA-knockdown effect of SHISA2 expression in PC-3 cells. Western blot analysis was performed using cells that had been transfected with shRNA-expressing vectors specific to SHISA2 and a negative control vector. ACTB was used as a loading control. (B) Colony formation assay of PC-3 cells transfected with the indicated shRNA-expressing vectors specific to SHISA2 and a negative control vector. Cells were visualized with 0.1% crystal violet staining after 14 days incubation with geneticin. (C) Cell viability assays of PC-3 cells transfected with the indicated shRNA-expressing vectors specific to SHISA2 and a negative control vector. Each mean value is plotted with error bars indicating the standard deviation. ACTB was used as a loading control. Experiments were performed in triplicate. *P<0.01, Tukey's test. SHISA2, shisa family member 2; siRNA, short interfering RNA; PC, prostate cancer; ACTB, β-actin; WB, western blotting; ABS, absorbance at 450 nm.
