TCC deletion in IFNAR1 decreases type I IFN signaling in human cells. HEK 293T cells were transfected with Cas9-expressing vector with or without endogenous IFNAR1 targeting gRNA. The endogenous IFNAR1-silenced (gRNA+) or control (gRNA−) cells were then transfected with WT or mutant IFNAR1 expression vectors together with an ISRE-driven luciferase reporter plasmid. a The luciferase activity was analyzed 18 h after IFN-α or IFN-β stimulation.
