The por1Δ mutation affects Snf1 nuclear enrichment. (A to C) Localization of Snf1-GFP was determined by fluorescence microscopy after growth to mid-log phase in selective SC containing 2% glucose (Glu) and after a 20-min shift to an otherwise identical medium containing 3% ethanol and 2% glycerol (EG) instead of glucose. The cells were imaged by differential interference contrast (DIC) microscopy. Nuclei were stained with DAPI. Scale bars, 5 μm.
