Mononuclear cells were separated from bone marrow samples of two MM patients (patient No1 and No2) using ficoll-paque solution. Percentage of myeloma cells in mononuclear fraction was more than 80 %. The primary myeloma cells form patients No.1 (A, B) or No. 2 (C, D) were incubated with 100 nM YM155 or ara-C 0.1 mM at 37°c for indicated time periods. Cell-cycle profiles were analyzed by flow cytometry using PI staining. The cell survival was determined by trypan blue dye exclusion assay.
